ABSTRACT

ObjectiveTo assess the effects of non-occupational mixed exposure to cadmium, lead and perfluoroalkyl substances (PFASs) on thyroid hormones (TH) in healthy adult residents in Shanghai. MethodsIn November 2018, adults in Shanghai Suburban Adult Cohort and Biobank study, who visited a community health service center for examination with no history of occupational exposure, thyroid diseases or chronic diseases, were recruited. A social-demographic information questionnaire survey was conducted and urine and blood samples were collected. Inductively coupled plasma mass spectrometry (ICP-MS) was used to measure their urinary cadmium (UCd), blood cadmium (BCd) and blood lead (BPb) concentrations. Ultra-high performance liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (UPLC-Q/TOF MS) was used to measure 11 kinds of PFASs. Total and free triiodothyronine (TT3, FT3), total and free thyroxine (TT4, FT4) and thyroid simulating hormones were measured by chemiluminescence enzyme-linked immunoassay. 436 participants were finally included and LASSO regression, multivariate regression and weighted quantile sum regression were used to evaluate the associations of these environmental pollutants with thyroid hormones. ResultsAmong the participants, 185 were male (42.5%) and the median age was 60 (P25‒P75: 50‒66). The detection rates of urinary cadmium, blood cadmium and blood lead were all more than 95% and the detection rates of 7 PFASs (PFOS, PFOA, PFHxS, PFUnDA, PFNA, PFD, and PFBS) were more than 90%. The median exposure level of PFOA was the highest (49.6 µg‧L-1) among PFASs, followed by PFHxS (22.8 µg‧L-1) and PFOS (15.4 µg‧L-1), and the median exposure levels of urinary cadmium, blood cadmium and blood lead were 0.7 μg‧g-1(Corrected for creatinine of urine), 0.8 µg‧L-1, and 15.4 µg‧L-1, respectively. The results showed that UCd was negatively associated with TSH and BCd was positively associated with TT3, while PFASs mainly affected FT4, TT4, and TT3, with gender differences. In males, 7 PFASs had a significant negative mixture effect on TT3 and TT4, while the direction of effect of PFASs in females differed, with PFOS and PFUnDA having a significant positive correlation with FT4 and TT4 while PFDA having a significant negative correlation with FT4 and TT4. ConclusionIn a healthy population with no occupational exposure, co-exposure to cadmium, lead and PFASs affects different thyroid hormone indicators, and such effect could be gender-related, indicating that the effect of mixed exposure to metal and emerging compounds on thyroid functions warrants further attention.

ABSTRACT

Objective:To prepare a dual targeting nanobubble both for HER2 target and cancer cell target, and compare the targeting ability with single targeting nanobubble either with HER2 targeting or cancer cell targeting ability in vitro. Methods:Nanobubbles(NBs) were prepared using the modified thin film hydration method and then connected with IR783 directly (NBs-IR783), HER2 antibody Affibody by avidin-biotin method (NBs-Affibody), or both of them (IR783-NBs-Affibody). The size distribution, stability, and biosafety of these contrast agents were observed. Flow cytometry and immunofluorescence were applied to compare the binding rate of these NBs against HER2 positive breast cancer cell in vitro. Results:The average size of NBs-Affibody, NBs-IR783 and IR783-NBs-Affibody was (538.4±95.8)nm, (551.8±114.8)nm, and (482.7±54.3)nm, respectively. The binding rate for HER2 positive cell was 26.6%, 97.6%, 84.5%, while for HER2 negative cell was 5.4%, 99.9% and 99.3%, respectively. The results of dual-labeled flow cytometry showed that the binding rate of IR783-NBs-Affibody mediated by IR783 to HER2 positive breast cancer cells were 79.5% and Affibody mediated HER2 targeting binding rate were 19.4%. However, NBs-IR783 only showed IR783-mediated tumor targeted binding without HER2 targeting ability with binding rate of 2.3%.Conclusions:The prepared IR783-NBs-Affibody has the dual targeting ability for HER2 positive breast cancer cell, which is superior to the single targeting NBs (NBs-Affibody and NBs-IR783). IR783-NBs-Affibody is optimal to meet the imaging requirements of tumor heterogeneity.

ABSTRACT

The morbidity of pancreatic ductal adenocarcinoma (PDAC) has been increasing over years, while the treatment efficacy and prognosis of PDAC remain far from satisfying. The newly-ermerged tumor immunotherapy has not only made lots of breakthroughs in various malignancies, but also brought an opportunity to the treatment of pancreatic cancer.PDAC immunotherapies, mainly including vaccine therapy, adoptive T cell thanfer therapy, checkpoint blockade therapy, have achieved a certain effect, however, the clinical outcomes have not been satisfactory. Therefore, the combination of immunotherapies based on different theoretical views is important and is likely to be the trend in the future. Carcinoma associated fibroblast (CAF) is the most common cell in pancreatic cancer stromal component. It will be helpful to develop more potential therapeutic targets by further exploring CAF and the mechanism of fibrosis mediated immunosuppression.

ABSTRACT

This study was aimed to observe the influence of water-solubility nipponica saponin on activation of TNF-α+IL-17-induced rat fibroblast-like synovial cell line RSC-364 cellular model nuclear transcription factor NF-κB pathway as well as TNF-α, IL-1, ICAM-1, MMP-2, MMP-3 secretion. IL-17+ TNF-α were used for stimulating RSC-364 to establish rheumatoid arthritis (RA) cellular model. Water-solubility nipponica saponin in different con-centrations was used for intervention. The influence of water-solubility nipponica saponin in different concentrations on cell viability was detected by semi-quantitative RT-PCR method. Changes in the level of TNF-α, IL-1, ICAM-1, MMP-2, and MMP-3 of culture supernatant were detected by ELISA. The results showed that the activation of NF-κB p65 in RSC-364 stimulated by TNF-α+ IL-17 can be inhibited by water-solubility nipponica saponin ac-cording to its concentration. It improved IκB-α expression, and inhibited TNF-α, IL-1, ICAM-1, MMP-2 and MMP-3 secretion. It was concluded that water-solubility nipponica saponin can inhibit the activation of NF-κB pathway, hinder the secretion and activation of multiple downstream genes, which may be its effect in inhibiting syn-ovial inflammation in RA.

ABSTRACT

[Objective] To explore the relativity between sPG and gastritis of H.pylori. [Method] Select 73 cases of chronic gastritis correctly diagnosed by gastric microscope and pathology, among which, 36 were chronic atrophic gastritis and 36 chronic superficial gastritis. 49 cases of positive H.pylori were administered with Lizhu Weisanlian tablets, measure the sPG before and after treatment. [Result] In the infected cases of H.pylori, PG Ⅰ was similar between atrophic gastritis and superficial gastritis; for PG Ⅱ value, atrophic gastritis group was more than superficial gastritis group, positive H.pylori patients higher than negative ones; PGR(PG Ⅰ/PG Ⅱ) was lower in atrophic gastritis group than superficial one, so was in positive H.pylori than negative one. After anti-H.pylori treatment, 87.75% patients were removed H.pylori, where the PG Ⅰ rose, PG Ⅱ lowered, PGR rose in atrophic gastritis group; in the superficial one where the H.pylori was not removed, no changes happened to PG Ⅰ, PG Ⅱ and PGR. In superficial gastritis group where the H.pylori was not removed, PGR rose. [Conclusion] H.pylori infection is correlated with sPG. Serum PG Ⅰ, PG Ⅱ and PGR have definite value in early diagnosing H.pylori concerned atrophic gastritis. PGR can be an index to evaluate the removal efficacy of H.pylori.
